Description
This SOP describes the procedure for the normalization of next-generation sequencing libraries for pooling and was developed by the National Aeronautics and Space Administration (NASA) GeneLab Sequencing Group (https://genelab.nasa.gov/genelab-sequencing-services/overview). Prior to pooling, the average insert size is confirmed to be comparable among the libraries using a TapeStation system, then the molarity of the libraries is calculated based on PicoGreen or Qubit quantification, and the fragment size is equalized. The SOP is a product of the GeneLab Sequencing Group and is available for download from https://genelab.nasa.gov/genelab-sequencing-services/sops and https://github.com/nasa/GeneLab-sampleProcessing.
