Description

This SOP describes the procedures for the homogenization of frozen tissues in lysis buffer (SOP #1.2) using a hand-held Polytron rotor stator homogenizer before nucleic acid isolation using Qiagen AllPrep Kits (SOP #3.1), Trizol reagent (SOP #3.2) or the MAXwell Feces Kit (SOP# 3.3). The procedure specified was developed by the National Aeronautics and Space Administration (NASA) GeneLab Sequencing Group (https://genelab.nasa.gov/genelab-sequencing-services/overview) and is used for mouse skin when larger volumes are required for lysis or in situations where the bead-based method has not been optimized.
