Description

This assay analytical validation and performance report describes the methods used to validate an automated singleplex immunohistochemical (IHC) staining procedure for the Bond Max instrument, which utilizes small reagent volumes (≥150 µl/slide), to assess CD3, CD4, CD45RO, CD57 (Leu-7), CD68, CD8, Foxp3, Granzyme B, PD-1, PD-L1 immunostaining in formalin-fixed, paraffin-embedded (FFPE) tumor and normal adjacent lung tissue, lymphocytes, and macrophages. The document also presents data that supports the reported accuracy, precision, analytical sensitivity and specificity, and reportable range for each antigen. It is used by the Cancer Immune Monitoring and Analysis Center (CIMAC) at MD Anderson Cancer Center (MDACC). It is a product of the CIMAC and Cancer Immunologic Data Center Network (CIMAC-CIDC Network; https://dctd.cancer.gov/ResearchNetworks/cimacs_cidc_network.htm), a Cancer Moonshot Initiative.
