Description

This analytical validation and performance report describes the methods used to validate the cytometry by time-of-flight (CyTOF) proteomics assay using whole blood collected from healthy donors into Smart tubes, which are analyzed on a CyTOF Helios 3 platform. The document also contains a list of intracellular and surface markers included on the CyTOF whole blood phosphophenotyping panel and presents data supporting the reported accuracy, inter- and intra-assay precision, analytical sensitivity and specificity, reference interval, failure rate, and turnaround time. It also includes an appendix that describes procedures for the cytokine-stimulated phosphoflow analysis of whole blood flu samples using CyTOF mass spectrometry. It is used by the Cancer Immune Monitoring and Analysis Center (CIMAC) at Stanford University. It is a product of the CIMAC and Cancer Immunologic Data Center Network (CIMAC-CIDC Network; https://dctd.cancer.gov/ResearchNetworks/cimacs_cidc_network.htm), a Cancer Moonshot Initiative.
