This document describes procedures for DNA and RNA extraction with the AllPrep DNA/RNA Mini kit from frozen melanoma and normal adjacent specimens, subsequent RNA purification with the mirVana miRNA Isolation kit, and melanin reduction of purified RNA with the cationic detergent cetyl- trimethylammonium bromide (CTAB).. It is a product of The Cancer Genome Atlas (TCGA) Program and was developed for use by Biospecimen Core Resource (BCR), located within the Biopathology Center (BPC) at Nationwide Children's Hospital in Columbus, Ohio.
