Purpose of Paper

The purpose of this paper was to determine the effects of refrigerated storage of red blood cells (RBC) on nitric oxide (NO) and methemoglobin formation in response to nitrite under deoxygenated and oxygenated conditions.

Conclusion of Paper

Under deoxygenated conditions, RBC refrigerated for 5-6 weeks had higher methemoglobin and NO formation 10 min after nitrite administration than those refrigerated for <1 week, but no difference was noted under normal oxygenation or after washing off the nitrite.

  1. Study Purpose

    The purpose of this study was to determine the effects of refrigerated storage of RBC on NO and methemoglobin formation in response to nitrite under deoxygenated and oxygenated conditions. Leukoreduced RBC from 6 patients were stored in saline-adenine-glucose-mannitol (SAGM) at 2-6 degrees C for <1 week or 5-6 weeks before being suspended in saline/glucose/Tris/sucrose/HCO3- and incubated at 37 degrees C. Cells were deoxygenated and sodium nitrite was added before measurement of NO for 10 min followed by methemoglobin levels.

    Summary of Findings:

    Under deoxygenated conditions, RBC refrigerated for 5-6 weeks had higher methemoglobin and NO formation 10 min after nitrite administration than RBCs refrigerated for <1 week (p=0.022 and p=0.026, respectively), but no difference was noted under normal oxygenation or after washing off the nitrite.
