A comparison of washed and volume-reduced platelets with respect to platelet activation, aggregation, and plasma protein removal.

Purpose of Paper
The purpose of this paper was to determine the effects of washing with normal saline (NSS) or Neutral calcium-free Ringer's acetate (NRA) and volume reduction on platelet (Plt) activation and aggregation.
Conclusion of Paper
Washing in NSS led to significantly increased Plt activation and mean fluorescence and decreased aggregation, mean Plt volume (MPV) and Plt distribution width (PDW) compared to volume reduced Plts. However, NSS washing removed 96% of the proteins present, while volume reduction only removed 51.1% of the proteins (p<0.001). Plt aggregation and activation and protein removal were comparable between NRA-washed and NSS-washed Plts, but Plt recovery and the change in PDW were lower among NRA-washed Plts than NSS-washed Plts.

Study Purpose
The purpose of this study was to determine the effects of washing Plts with NSS or NRA and volume reduction on Plt activation and aggregation. Apheresis Plts that were less than 12 h past expiration were split for washing and volume reduction. After volume reduction, Plts were resuspended in plasma. After washing or volume reduction, Plts were allowed to rest for 60 min, after which point they were agitated for 15 min before sampling.
Summary of Findings:
Washing Plts in NSS led to significantly increased Plt activation, as measured by the percentage of P-selectin positive Plts and mean fluorescence, compared to volume-reduced Plts (p=0.001 and p=0.004, respectively). The NSS-washed Plts displayed decreased aggregation compared to volume-reduced Plts (p=0.004). Further, the MPV and PDW were lower in NSS-washed Plts than volume-reduced Plts. However, NSS washing removed 96% of the proteins present, while volume reduction only removed 51.1% of the proteins (p<0.001). Plt recovery was comparable between NSS-washed and volume-reduced Plts. Aggregation and activation were comparable between NRA-washed and NSS-washed Plts, but Plt recovery and the change in PDW were lower among NRA-washed Plts than NSS-washed Plts (p=0.008 and p=0.002, respectively).
