Study Purpose
The purpose of this study was to determine the effects of post-thaw dilution and washing and post-thaw storage at room temperature on cord blood cell recovery and viability. Liquid nitrogen frozen cord blood units were thawed in a water bath at 37 degrees C and split into three aliquots. One aliquot was left unwashed while the other two were diluted with albumin dextran. One of the albumin dextran diluted aliquots was further processed by centrifugation and removal of the supernatant (washed) while the other was subjected to no further processing (dilution).
Summary of Findings:
TNC recovery was generally similar regardless of post-thaw processing methods, but after 0 and 48 h of post-thaw storage at room temperature, TNC recovery was significantly lower when cells were washed with albumin dextran than when they were not washed. CD34 recovery declined with post-thaw storage at room temperature of unwashed, undiluted specimens with significance at 24 h compared to 0 h (p<0.01), but CD34 recovery remained relatively constant in washed or diluted specimens. The number of CFCs declined with post-thaw storage at room temperature, and the decline was more rapid in unwashed, undiluted specimens than washed or diluted specimens (p<0.01). Similarly, viability decreased with increasing post-thaw storage at room temperature and was lower in unwashed, undiluted specimens than in washed or diluted specimens after 24, 32 and 48 h (p<0.05, p<0.05, and p<0.01, respectively).
