Purpose of Paper

The purpose of this paper was to determine the effects of freezing and post-thaw refrigerated storage of plasma on clotting factor activity.

Conclusion of Paper

Compared to fresh plasma, plasma that had previously been frozen had lower levels of fibrinogen, factor (F)II, FV, FVII, FX, FXIII, protein C (PC) and antithrombin III  (ATIII). FII, FV, FVII, FVIII, FIX, FX, FXI and free protein S (FPS) decreased significantly between 0 h and 6 days of post-thaw refrigerated storage. Further, compared to levels measured immediately after thawing, previously frozen plasma stored for 6 days had higher FXII, FXIII and PC.

  1. Study Purpose

    The purpose of this study was to determine the effects of freezing and post-thaw refrigerated storage of plasma on clotting factor activity. Sodium citrate plasma from 20 healthy donors was stored frozen at -30°C for at least 8 weeks before thawing in a Plasmatherm III. Thawed plasma was stored at 4°C.

    Summary of Findings:

    Compared to fresh plasma, plasma that had previously been frozen had lower levels of fibrinogen, FII, FV, FVII, FX, FXIII, PC and ATIII (p<0.001, all).  FII, FV, FVII, FVIII, FIX, FX, FXI and FPS decreased significantly between 0 h and 6 days of post-thaw refrigerated storage  (p<0.001, all). Further, compared to levels of activity measured immediately after thawing, previously frozen plasma stored for 6 days had higher FXII (p=0.023), FXIII (p<0.001) and PC (p=0.003). Levels of von Willebrand factor antigen (VWF-Ag) and FPS were not affected by freezing or post-thaw refrigerated storage of plasma.
