Purpose of Paper

The purpose of this paper was to assess the stability of urinary proteins after storage at -20, 4, or 22 degrees C for varying durations in the presence or absence of a preservative.

Conclusion of Paper

Of the seven urinary proteins examined, alpha1-antitrypsin alone decreased after storage at 22 or 4 degrees C for 7 or 30 d, respectively. Frozen storage or urine specimens with or without a preservative adversely affected the levels of urinary protein after 30 d or longer.

Studies

  1. Study Purpose

The purpose of this paper was to assess the stability of urinary proteins after storage at -20 (1-180 d), 4 (1-30 d), or 22 (1-7 d) degrees C in the presence or absence of a preservative that contained benzamidinium chloride, EDTA, tris (hydroxymethyl)-aminomethane, and azide.

    Summary of Findings:

Concentrations of urinary proteins were comparable when specimens were stored at 22 or 4 degrees C for up to 7 or 30 d, respectively, with or without a preservative, with the exception of alpha1-antitrypsin which decreased by approximately 35% after storage in preservative-free specimens for the maximum duration (7 d at 22 degrees C, 30 d at 4 degrees C). Urine specimens stored at -20 degrees C for 180 d differed in IgG and alpha1-antitrypsin levels or lambda-immunoreactivity and alpha1-antitrypsin levels in preservative-treated and preservative-free specimens, respectively, compared to immediately processed controls. Alpha1-antitrypsin and IgG levels differed significantly from controls after 30 d or more of frozen storage.
