Purpose of Paper

The purpose of this paper was to compare the yield of cell-free fetal DNA (cffDNA) and total cell-free DNA from plasma extracted using three different methods.

Conclusion of Paper

Use of either the QIAamp DSP Virus Kit or the NucliSens Magnetic Extraction system resulted in significantly higher yields of cffDNA and total cell-free DNA than extraction with the QIAamp blood kit.

  1. Study Purpose

    The purpose of this study was to compare the yield of cffDNA and total cell-free DNA from plasma using three different extraction methods. DNA was extracted from frozen (-30ºC) plasma of 75 Rhesus blood group D (RhD)-negative pregnant women who gave birth to RhD-positive infants. cffDNA and total cell-free DNA were quantified through real-time PCR amplification of RhD and glyceraldehyde 3-phosphate dehydrogenase (GAPDH), respectively.

    Summary of Findings:

    Extraction of cffDNA with the QIAamp DSP Virus Kit or the NucliSens Magnetic Extraction system resulted in 2.4- and 1.7-fold higher cffDNA yields per mL of plasma  in comparison to the QIAamp blood kit (p=0.00007 and p=0.00001, respectively). Further, yields of non-fetal cell free DNA per mL of plasma were 2.1- and 2.3-fold higher using the QIAamp DSP Virus Kit or the NucliSens Magnetic Extraction system, respectively, than the QIAamp blood kit (p=0.002 and p=0.039, respectively).
