Purpose of Paper

The purpose of this paper was to compare miRNA levels in formalin-fixed paraffin-embedded (FFPE) surgical specimens with those in fine needle aspirates (FNA) which were snap-frozen in lysis buffer.

Conclusion of Paper

The RNA yields from snap-frozen FNA were lower than those from FFPE surgical specimens. Further, the cycle threshold (CT) values for amplification of U6 using RNA from snap-frozen FNA were higher than those obtained using RNA from FFPE surgical specimens.

  1. Study Purpose

    The purpose of this study was to compare miRNA levels in FFPE surgical specimens with those in FNA which were snap-frozen in lysis buffer. RNA was extracted from tumor regions of 2-4 unstained sections using RecoverAll Total Nucleic Acid Isolation kit (Ambion). FNA were suspended in lysis buffer (Qiagen), snap frozen, and stored at -80 degrees C until RNA extraction using mirVana lysis buffer and phenol-chloroform extraction with columns.

    Summary of Findings:

    The RNA yields from snap-frozen FNA were lower than those from FFPE surgical specimens (0.41-12 ug versus 1.1-27 ug). The range of absorbance at 260/280 nm was larger for RNA from snap-frozen FNA than from FFPE surgical specimens (1.61-2.82 verses 1.90-2.12). Further, the CT values for amplification of U6 using RNA from snap-frozen FNA were higher than those obtained using RNA from FFPE surgical specimens. Patient diagnosis had a significant effect on the levels of miR-146b, -221, and -222.
