Purpose of Paper

The purpose of this paper was to compare tissues preserved in universal molecular fixative (UMFIX) and then paraffin embedded with formalin fixed, paraffin-embedded (FFPE) and fresh frozen tissues.

Conclusion of Paper

The authors report that UMFIX adequately preserved histology (similar to FFPE tissues), RNA quality and quantity (similar to fresh-frozen tissues), and protein immunoreactivity (similar to FFPE tissues) when preserved immediately after excision; however RNA quality and quantity were adversely affected by delays in fixation (as short as 5 min), or failure to maintain an RNase-free environment.

    The purpose of this study was to compare tissues preserved in universal molecular fixative (UMFIX) and then paraffin embedded with formalin fixed, paraffin-embedded (FFPE) and fresh frozen tissues. Adrenal, breast, colon, eye, esophagus, kidney, liver, lung, lymph node, skeletal muscle, pancreas, parathyroid, parotid, prostate, skin, small intestine, soft tissue, spleen, thyroid, tonsil, and uterus were investigated.

    Summary of Findings:

    The authors report that UMFIX adequately preserved histology (similar to FFPE tissues), RNA quality and quantity (similar to fresh-frozen tissues), and protein immunoreactivity (similar to FFPE tissues) when preserved immediately after excision; however RNA quality and quantity were adversely affected by delays in fixation with unpredictable RNA quality observed with a delay of 5 to 30 minutes, or failure to maintain an RNase-free environment. UMFIX-fixed paraffin embedded tissue stored at room temperature for 8 weeks yielded intact RNA.
