Purpose of Paper

The purpose of this paper was to determine the effects of delayed processing, processing temperature and storage after processing on the proteome profiles of serum.

Conclusion of Paper

When serum was stored at room temperature for 4 h prior to protein extraction, all peaks over the 4000 m/z range were absent, but the spectra of serum did not change significantly during 3 weeks of storage of processed samples at -20 degrees C. The profiles obtained were similar when sample preparation was done at 4 or 21 degrees C, but profiles were different and there was more noise when preparation was performed at 30 degrees C. A very different profile was obtained from a single patient with cancer compared to a single normal control, but the authors report this to be unusual.

Studies

  1. Study Purpose

The purpose of this study was to determine the effects of delayed processing, processing temperature and storage of processed samples at -20 degrees C on the proteome profile of serum. Specimens were frozen at -80 degrees C and thawed up to three times including thawing for this study.

    Summary of Findings:

When serum was stored at room temperature for 4 h prior to protein extraction, all peaks over the 4000 m/z range were absent, but the spectra of serum did not change significantly after 1 or 3 weeks of storage of processed samples at -20 degrees C compared to samples analyzed immediately after processing. The profiles obtained were similar when sample preparation was done at 4 or 21 degrees C, but profiles were different and there was more noise when preparation was performed at 30 degrees C. A very different profile was obtained from a single patient with cancer compared to a single normal control, but the authors report this to be unusual.
