Study Purpose
The purpose of this study was to conduct a comparative analysis of tumor marker protein expression (E-cadherin, HER2, beta-actin, alpha-tubulin, GAPDH, snail, and a mutant E-cadherin with an exon 9 deletion) after protein extraction from FFPE and fresh frozen tissues, which included placenta, colon, gastric, breast, and pancreatic tissue.
Summary of Findings:
Quantities of full length immunoreactive proteins extracted from FFPE tissue were comparable to fresh frozen controls. While protein patterns from FFPE tissue were similar they were not identical to fresh frozen controls. All tumor markers examined (E-cadherin, HER2, beta-actin, alpha-tubulin, GAPDH, snail, and a mutant E-cadherin with an exon 9 deletion) were observed at comparable levels in FFPE and fresh frozen tissues as measured by Western blot analysis and reverse phase microarray.
