Purpose of Paper

A comparative analysis of protein quantity, quality, and tumor marker abundance among routine formalin-fixed paraffin embedded (FFPE) and fresh frozen tissues was performed.

Conclusion of Paper

No differences were observed in the relative abundance of tumor marker proteins, which included membrane, cytoplasmic, and nuclear proteins, among FFPE and fresh frozen tissue. While protein patterns were very similar between fixed and frozen tissue, the patterns were not identical.

Studies

  1. Study Purpose

The purpose of this study was to conduct a comparative analysis of tumor marker protein expression (E-cadherin, HER2, beta-actin, alpha-tubulin, GAPDH, snail, and a mutant E-cadherin with an exon 9 deletion) after protein extraction from FFPE and fresh frozen tissues, which included placenta, colon, gastric, breast, and pancreatic tissue.

    Summary of Findings:

Quantities of full length immunoreactive proteins extracted from FFPE tissue were comparable to fresh frozen controls. While protein patterns from FFPE tissue were similar they were not identical to fresh frozen controls. All tumor markers examined (E-cadherin, HER2, beta-actin, alpha-tubulin, GAPDH, snail, and a mutant E-cadherin with an exon 9 deletion) were observed at comparable levels in FFPE and fresh frozen tissues as measured by Western blot analysis and reverse phase microarray.
