Study Purpose
The purpose of this study was to compare protein quality and profiles of FFPE and case-matched frozen glioblastoma specimens after manual dissection, detergent-based protein extraction, and heat-induced antigen retrieval.
Summary of Findings:
Snap-frozen and manually microdissected glioblastoma specimens yielded protein of superior quality compared to case-matched FFPE specimens. Heat-induced antigen retrieval combined with the highly sensitive separation strategy and mass spectrometry enabled the researchers to identify similar proteins in FFPE and fresh-frozen tissue. Of the 3902 proteins identified only 8.5% were unique to FFPE specimens, with an approximate 88% overlap among fixation methods.
