Study Purpose
Comparative analysis of immunohistochemical staining for several nuclear markers, (minichromosome maintenance protein 2 (MCM-2), p27, Ki-67), membrane markers (c-erb-B2), and CD-34 and alpha-methylacyl-CoA racemase (AMACR) in prostate biopsies fixed in either Bouins or formalin and paraffin embedded.
Summary of Findings:
While FFPE tissue displayed appropriate MCM-2 immunostaining, staining in BFPE tissue was negative and was unaltered by shortened fixation, lengthened primary antibody incubation, lengthened antigen retrieval, or methanol postfixation. P27 immunostaining was significantly reduced in BFPE compared to FFPE tissues, and included extensive nonspecific cytoplasmic staining that was minimal in FFPE samples. In BFPE samples, c-erb-B2 staining was strong and inappropriate in the majority of samples examined. However, immunostaining for AMACR, Ki-67, and CD-34 was comparable among BFPE and FFPE samples.
