Study Purpose
The purpose of this study was to determine the effects of leukoreduction filtration on HHV-8 viral load in blood and to compare HHV-8 load in blood and plasma from HIV positive men with and without KS. Blood was collected in acid-citrate-dextrose (ACD), frozen at -80 degrees C, and transported the same day or the next day, but the transport method and distance were not specified. Once in the laboratory, an aliquot of blood was removed to obtain plasma, another was removed for assay, and the remaining blood was leukoreduced. The method used to determine CD4 count was not specified.
Summary of Findings:
Before leukofiltration, 12 of 21 blood specimens from patients with KS had detectable HHV-8, but after leukofiltration, only 5 of 21 specimens from patients with KS had detectable HHV-8. Further, the HHV-8 levels were more than 10 fold lower after leukofiltration of blood than before leukofiltration. Plasma levels of HHV-8 were generally comparable to levels in leukofiltered blood and were 1-20% of those in unfiltered blood. In the 12 patients with detectable HHV-8 in blood, there was an inverse relationship between HHV-8 viral load and CD4 count (Spearman's rank coefficient, -0.615; p = 0.03). HHV-8 was not detectable in the blood of patients without KS.
