Preanalytical stability of 17 analytes in whole blood during transportation.

Purpose of Paper

This paper investigated the stability of 17 analytes in whole blood transported and stored at room temperature for up to 10 h before serum separation.

Conclusion of Paper

For almost every analyte evaluated, levels outside of the reference range were found in one or more specimens. When serum separation was delayed by 10 h, levels of Apolipoprotein B (APOB) and Insulin-like growth factor 1 (IGF-1) increased (P=0.0380 and P=0.0102, respectively) and growth hormone (GH) and Thyroxin binding globulin (TBG) decreased (P=0.000 and P=0.0018, respectively). While statistically significant, the changes in APOB, IGF-1, GH and TBG were small and within the limits of the pre-established analytical variation.

  1. Study Purpose

    This study investigated the stability of 17 analytes in whole blood transported and stored at room temperature for up to 10 h before serum separation.  Blood was collected from ten outpatients (diagnosis not specified) into three Bio-One serum vacutainer tubes. One tube from each patient was stored upright at room temperature for between 1 h 5 min to 1 h 45 min, before centrifugation (baseline) while the other two tubes were stored vertically for 4 and 8 h, respectively, and then transported at room temperature (1.5 h) before serum separation (6 h 5 min-6 h 45 min and 10 h 5 min-10 h 45 min after collection, respectively).  Serum was separated by centrifugation at 2654 g for 5 min and stored at -20°C until analysis. Levels of lipoprotein a, immunoglobulin G1, β-2-Microglobulin, κ free light chain, and λ free light chain were analyzed immunoturbidmetrically using an Optilite instrument. Levels of peptidyl peptidase A and total protein were quantified using a Pentra C400 spectrophotometer.  Apolipoprotein B (ApoB) levels were quantified by nephelometry. Insulin-like growth factor 1 (IGF-1) and growth hormone were measured using iSys immunoassays. Thyroxin binding globulin was quantified by radioimmunoassay. Anticardiolipin antibodies IgM, prolactin, thyroid peroxidase antibodies, thyroglobulin, and thyroglobulin antibodies were measured using a KRYPTOR Instrument, and 17-Hydroxyprogesterone was quantified by mass spectrometry.

    Summary of Findings:
