Purpose of Paper

The purpose of this paper was to determine the effects of storage duration on microRNA (miRNA) expression in formalin-fixed paraffin-embedded (FFPE) tumor blocks.

Conclusion of Paper

miRNA expression of the lung cancer specimens showed a high degree of correlation regardless of storage duration.

  1. Study Purpose

    The purpose of this study was to determine the effects of storage of lung tumor FFPE blocks and tumor tissue origin on miRNA expression in 333 FFPE tumor blocks from 22 different tissue types. RNA was obtained from 7-10 10 uM sections by xylene deparaffinization, extraction with the miRdictor kit, and miRNA profile was determined using custom miRdicator arrays. The ability of the 48 identified miRNA to function as tumor classifiers was tested by blinded analysis of 83 FFPE tumor blocks.

    Summary of Findings:

    Overall miRNA expression of any one lung cancer specimen stored for 2-11 years showed a correlation of 0.9 with the median expression of the 13 specimens. The miRNA profile of most cancers was similar between primary and metastasis, but this was not true for stomach cancers where 3 miRNAs were differentially expressed. 48 miRNAs were identified as tumor classifiers and validated using real-time PCR.
