Purpose of Paper

The purpose of this paper was to assess effects of periods of cold ischemia on RNA quality and mRNA expression.

Conclusion of Paper

Although RNA quality was unaffected by periods of cold ischemia up to 60 min, mRNA expression was significantly affected in >50% of the genes analyzed by microarray. Expression of the majority of these genes was significantly altered after periods of ischemia >20 min, and included robust and diverse changes in expression (0 to 20-fold). However, Northern blot analysis of GADPH expression yielded no significant alterations with varying ischemia durations.

    RNA quality, assessed by gel electrophoresis and visual examination of 28S and 18S ribosomal bands, was unaffected by ischemia periods of up to 60 min.

    To determine the effects of varying durations of cold ischemia (5 to 60 min at room temperature post-excision) on mRNA expression as measured by microarray.

    Summary of Findings:

    Of the 2400 genes assessed by microarray, mRNA expression of 1420 genes (59%) were significantly affected in a time-dependent manner. Magnitude of ischemia-dependent changes in gene expression ranged from 0 to 20-fold. Further, the majority of genes exhibiting >2-fold changes in expression occurred after 20 min or more of cold ischemia time.
