Purpose of Paper

The purpose of this paper was to determine the effects of room temperature cold ischemia time on proliferating cell nuclear antigen (PCNA) immunohistochemical (IHC) staining.

Conclusion of Paper

While fixation delays had no significant effect on PCNA grade (low versus high), fixation delays of 24 h resulted in a 10% average increase in PCNA index (percentage of positive tumor cell nuclei). The authors note there was marked intertumoral heterogeneity with respect to PCNA indices for each cold ischemia time.

    The purpose of this study was to determine the effects of room temperature (20 degrees C) cold ischemia time on PCNA IHC staining in formaldehyde-fixed specimens.

    Summary of Findings:

    While fixation delays had no significant effect on PCNA grade (low: less than 50% positive nuclei versus high: greater than 50% positive nuclei), fixation delays of 24 h resulted in a 10% average increase in PCNA index.
