Purpose of Paper

The purpose of this paper was to determine the effects of hemolysis and assay incubation time on the measurement of insulin in serum by radioimmunoassay (RIA).

Conclusion of Paper

The measured level of insulin increased linearly with increasing hemolysis, but the degree of increase also depended on the incubation time of the RIA.

  1. Study Purpose

    The purpose of this study was to determine the effects of hemolysis and assay incubation time on the measurement of insulin in serum by RIA. Blood specimens were obtained from 3 volunteers. Hemolysis was induced in the blood specimens by passing the blood through a 22 gauge needle before centrifugation. Serum from hemolyzed blood was added at increasing amounts to non-hemolyzed specimens.

    Summary of Findings:

    The measured level of insulin increased linearly with increasing hemolysis. A corresponding decrease in the insulin tracer solution was noted with increasing hemolysis. When the incubation time used with the Micromedic RIA was reduced from 16 h to 90 min (incubation time for the Immunophase RIA), the effect of hemolysis on insulin levels was partially attenuated.
