Purpose of Paper

The purpose of this paper was to determine the effects of anticoagulant on the stability of adiponectin in blood during storage on ice for 36 h and the intraindividual variability in adiponectin levels.

Conclusion of Paper

Adiponectin concentrations were stable in sodium heparin specimens during the 36 h of storage and in EDTA specimens for 24 h, but concentrations increased in EDTA specimens stored on ice for 36 h compared to specimens analyzed immediately. Average adiponectin concentrations decreased slightly over the course of 1 year, but when corrected for body mass index (BMI), the change was insignificant.

Studies

  1. Study Purpose

The purpose of this study was to determine the stability of adiponectin in blood during storage on ice for 36 h with EDTA or sodium heparin and the intraindividual variability in adiponectin levels. Blood was collected after a 12 h fast from 6 males and 6 females into EDTA tubes and from 12 females into sodium heparin tubes. After storage, specimens were centrifuged, aliquoted and plasma was stored in liquid nitrogen. To determine the intraindividual variability, 2 specimens were collected 1 year apart from 20 men.

    Summary of Findings:

Adiponectin concentrations were stable in sodium heparin specimens during the 36 h of storage and in EDTA specimens for 24 h, but concentrations increased by an average of 2.63 mg/L in EDTA specimens stored on ice for 36 h compared to specimens analyzed immediately (p=0.05). However, the authors report that the effect of storing EDTA blood on ice was no longer significant when specimens were stratified by gender. Intrassay variability was 16% for EDTA specimens and 26% for sodium-heparin specimens. Adiponectin concentrations in individuals decreased slightly over the course of 1 year, but when corrected for BMI, the change was insignificant. Adiponectin concentrations were inversely correlated with BMI in 2000 (r=-0.47, p=0.04) and 2001 (r=-0.45, p=0.04). The authors report no correlation of volunteer age with adiponectin levels.
