Purpose of Paper

The purpose of this paper was to determine the effects of centrifuge speed and duration on coagulation and hematological parameters in automated and manually processed plasma.

Conclusion of Paper

Plasma specimens obtained by manual centrifugation at 1500 x g for 20 min or by automated centrifugation at 3000 x g for 7 min were each classified as platelet poor plasma based on platelet counts, but the platelet count was much higher in plasma processed manually at 1500 x g for 20 min. Generally, levels of coagulation factors were strongly or very strongly correlated between plasma obtained by automated centrifugation at 3000 x g for 7 min and plasma obtained by manual centrifugation at 1500 x g for 5 or 20 min, but correlations in levels of Factor (F)VII and FX in plasma obtained through automated centrifugation and plasma obtained by manual centrifugation at 1500 x g for 5 min were strong.

  1. Study Purpose

    The purpose of this study was to determine the effects of centrifuge speed and duration on coagulation and hematological parameters in automated and manually processed plasma. Specimens were centrifuged automatically at 3000 x g for 7 min or manually at 1500 x g for 5 min or 20 min.  Sodium citrate blood was processed within 2-3 h of collection.

    Summary of Findings:

    Platelet counts decreased from 17,000-125,000 platelets/uL in specimens centrifuged manually at 1500 x g for 5 min, to 3000-9000 platelets/uL in plasma after automated centrifugation at 3000 x g for 7 min and to 1000-6000 platelets/uL in plasma processed manually at 1500 x g for 20 min. Thus the plasma obtained by the automated centrifugation or centrifugation at 1500 x g for 20 min was classified as platelet poor plasma while the plasma obtained by manual centrifugation at 1500 x g for 5 min was not. When specimens were processed with automated centrifugation at 3000 x g for 7 min, levels of fibrinogen, FVIII, FIX, FXI, FXII and D-dimer, partial thromboplastin time (PTT), the international normalized ratio (INR), and quick activated partial thromboplastin time(aPTT) were very strongly correlated (R2=0.964-0.987), FV levels were strongly correlated (R2=0.888), and FVII and FX were modestly correlated (R2=0.586-0.685) with those in plasma obtained by manual centrifugation at 1500 x g for 5 min. Very strong correlations in levels of FV, FVIII, FIX, FXI, and FXII (R2=0.916-0.980) and strong correlations in levels of FVII and FX (R2=0.777-0.822) were observed between plasma processed by automated centrifugation at 3000 x g for 7 min and plasma obtained by manual centrifugation at 1500 x g for 20 min. Levels of FV, FVII, FVIII, FIX, FXI and FXII were very strongly correlated (R2=0.917-0.986) and levels of FX were strongly correlated (R2=0.792) between plasma obtained by manual centrifugation at 1500 x g for 5 min and 30 min.
