Evaluation of BD Vacutainer® Barricor™ blood collection tubes for routine chemistry testing on a Roche Cobas® 8000 Platform.

Purpose of Paper

This paper compared levels of 50 clinical chemistry analytes as well as residual cell counts in plasma from blood collected in PST II and Barricor tubes. The stability of five analytes in plasma from PST II and Barricor tubes during refrigerated storage was also investigated.

Conclusion of Paper

Levels of all 50 analytes were comparable in plasma from PSTII and Barricor tubes, but residual platelet counts were higher in plasma from PSTII tubes. The stability of aspartate aminotransferase (AST), potassium (K+), phosphate, lactate dehydrogenase (LDH), and glucose levelswas greater in plasma from Barricor tubes than PSTII tubes during refrigerated storage.

  1. Study Purpose

    This study compared levels of 50 clinical chemistry analytes as well as residual cell counts in plasma from blood collected in PST II and Barricor tubes. The stability of five analytes in refrigerated plasma from PST II and Barricor tubes was also investigated. Blood was collected from 150 patients from the emergency department and outpatient clinics into Barricor and PST II tubes.  Within 2 h of collection, plasma was obtained from blood collected in Barricor tubes by centrifugation at 4000 x g for 3 min and from the PST II tubes by centrifugation at 1300 x g for 10 min. Levels of 50 clinical chemistry analytes were determined using a Cobas 8000. Cellular contamination was evaluated in specimens from 20 participants using a DxH 800 hematology autoanalyzer.  The stability of AST, glucose, potassium, phosphate, and LDH were determined by daily analysis of four plasma specimens stored at 4˚C over a 10-day period.

    Summary of Findings:

    The levels of all 50 analytes were comparable across the concentration range between plasma from Barricor and PST II tubes, but there was a very narrow range of prostate specific antigen (PSA and free PSA values) in this study. The percent biases ranged from -1.5% (creatinine and ALT) to 6.1% (direct bilirubin) and all values were strongly correlated between tube types (R≥0.9144).  Average platelet counts were higher in plasma from PST II tubes than Barricor tubes (P<0.05). Red blood cell (RBC) and white blood cell (WBC) counts were below reporting limits, regardless of tube type

    Refrigerated storage of plasma from PST II tubes resulted in clinically significant (greater than 10%) mean increases in AST by day 6, K+ by day 4, phosphate and LDH by day 3, and and greater than a 10% decrease in glucose by day 4. In contrast, levels of AST, potassium, glucose, and LDH were stable (<10% change) over the 10 days of storage of plasma from Barricor tubes and clinically significant changes in phosphate were observed only after day 5.
