    The purpose of this study was to determine the effect of storage of saliva at -20 degrees C for 11 years on DNA integrity.

    Summary of Findings:

    Long term frozen storage of saliva saturated cotton wads did not inhibit the ability to extract DNA from buccal cells suitable for PCR-based genotyping. DNA yields, as determined by spectrophotometer, were variable and seemed to correlate with the amount of saliva contained in the spit wad. Average yield was 17.3 +/- 11.9 ug. Average quality as measured by A260/A280 ratio was 1.937 +/- 0.226 prior to concentration. PCR amplification of a functional polymorphism in the serotonin transporter gene promoter region was successful for 98.3% of the 783 samples.
