Purpose of Paper

The purpose of this paper was to determine the effects of storage at 4°C or room temperature, before or after centrifugation, and agitation prior to centrifugation on fibrinogen levels, D-dimer levels, thrombin time (TT), prothrombin time (PT) and activated partial thromboplastin time (APTT) in plasma.

Conclusion of Paper

Statistically significant changes in levels of all analytes were observed when specimens were stored for 2 h or more at 4°C or room temperature before or after centrifugation, regardless of agitation before centrifugation. However, clinically relevant changes (>10%) were only observed for APTT. Clinically relevant changes in individual APTT measurements were observed in >25% of specimens stored for 24 h, with or without agitation, at 4°C or room temperature, before or after centrifugation, but clinically relevant changes in mean APTT measurements were only observed when plasma was stored for 24 h at either temperature after centrifugation.

  1. Study Purpose

    The purpose of this study was to determine the effects of storage at 4°C or room temperature, before or after centrifugation, and agitation prior to centrifugation on fibrinogen levels, D-dimer levels, TT, PT and APTT in plasma. Sodium citrate blood from 120 Hepatitis B positive patients was collected after an overnight fast. Blood was stored with or without mechanical agitation and then centrifuged at 300 x g for 10 min to obtain plasma or was centrifuged immediately and stored as plasma. For each experimental temperature/condition, 20 specimens were used. Clinical relevance was defined as >10% change in value. The change was further characterized as moderate or large if observed in <25% or ≥25% of specimens, respectively.

    Summary of Findings:

    Statistically significant changes in levels of all analytes were observed when specimens were stored for 2 h at 4°C or room temperature before or after centrifugation, regardless of agitation prior to centrifugation. However, with the exception of APTT, none of the analytes displayed a >10% change in mean levels. Average APTT measurements increased by 13.55% and 10.52% when plasma was stored at 4°C or room temperature, respectively, for 24 h, but average APTT measurements remained within 10% of baseline when centrifugation was delayed by 24 h, regardless of agitation during storage. Further, more than 25% of specimens had a >10% change in APTT when stored for 24 h at room temperature or 4°C before or after centrifugation, regardless of whether they were agitated before centrifugation.
