This paper investigated the effects of storing whole blood at room temperature and 4-8˚C in Roche Cell-Free DNA Collection Tubes on cell-free DNA (cfDNA) concentrations in plasma.

Conclusion of Paper

cfDNA levels in plasma were not significantly affected by storage of whole blood in Roche Cell-Free DNA tubes for 24 or 48 h at either room temperature or 4-8˚C. The authors report visible hemolysis in all specimens stored for 48 h at 4-8˚C.

  1. Study Purpose

    This study investigated the effects of storing whole blood at room temperature and 4-8˚C in Roche Cell-Free DNA Collection Tubes on cfDNA concentrations in plasma. Whole blood was collected from 30 healthy individuals into five Roche Cell-Free DNA Collection Tubes. Tubes were centrifuged immediately, after 24 or 48 h at room temperature, or after 24 or 48 h at 4-8˚C. After storage, plasma was obtained by centrifugation at 1600 x g for 15 min. cfDNA was extracted from 2 mL of plasma using the QIAamp Circulating Nucleic Acid Kit using a QIAsymphony. cfDNA was stored at -60˚C until analysis. cfDNA was quantified by amplification of a 110 bp fragment of β-globin using EvaGreen-based Droplet Digital PCR.

    Summary of Findings:

    cfDNA levels in plasma were not significantly affected by storage of whole blood in Roche Cell-Free DNA Tubes for 24 or 48 h at either room temperature or 4-8˚C. The authors report visible hemolysis in all specimens stored for 48 h at 4-8˚C.
