  1. Study Purpose

    This study explored how circadian rhythm may influence the number of circulating tumor cells (CTCs) detected by the CellSearch method in peripheral blood of metastatic breast cancer patients. Blood (7.5 mL) was collected in CellSave tubes at 8:00 a.m. and 8:00 p.m. of the same day from 51 patients with metastatic breast cancer and processed using the CellSearch method within 72 h. Control specimens were collected at 8:00 a.m. from healthy, age-matched volunteers and run in parallel.

    Summary of Findings:

    No significant differences in numbers of CTCs in metastatic breast cancer patient specimens were observed between case-matched peripheral blood specimens collected 12 hours apart (P=0.8427). Prognostic classification was unchanged in blood specimens collected 12 hours apart in 96% of patients when a threshold of ≥5 CTCs/7.5mL was applied, as only two of the 51 patients were reclassified based on the time of CTC collection (patient 1: 0 vs. 10 CTCs; patient 2: 8 vs. 3 CTCs). No CTCs were isolated from healthy control specimens.
