Purpose of Paper

The purpose of this paper was to compare B-type natriuretic peptide (BNP) levels in plasma collected in EDTA polyethylene terephthalate (PET) tubes, EDTA glass tubes, and lithium heparin glass tubes and to investigate the post-centrifugation stability of BNP in each tube type during 4 h of room temperature storage.

BNP levels were significantly higher in plasma collected in EDTA PET tubes compared to EDTA glass tubes, and were higher in EDTA glass tubes compared to lithium heparin glass tubes. BNP levels were unaffected by post-centrifugation storage at room temperature of plasma  in EDTA/PET tubes, but levels decreased slightly in plasma collected and stored in EDTA/glass tubes. BNP levels decreased progressively when plasma was stored in lithium heparin glass tubes after centrifugation. When 2000 ng/L BNP was added to the plasma of healthy volunteers, 86.04% was recovered from the EDTA glass tube, 88.23% from the EDTA PET tube, but only 77.46% from the lithium heparin glass tube.

  1. Study Purpose

    The purpose of this study was to compare BNP levels in plasma collected in EDTA PET tubes, EDTA glass tubes and lithium heparin glass tubes and to investigate the post-centrifugation stability of BNP in each tube type during 4 h of room temperature storage. Blood was collected after overnight fasting from 36 individuals being treated as in-patients (16 females and 20 males with a BNP levels between 10-5005 ng/µL) into lithium heparin glass, EDTA glass, and EDTA/PET tubes. Plasma was obtained by centrifugation at 2000 g for 10 min and BNP levels were measured after 0, 0.5, 1, 2, and 4 h at room temperature. BNP was quantified using a sandwich enzymatic chemiluminescent assay on a CL-6000i automatic chemiluminescence immunoanalyzer.  To evaluate the accuracy of the BNP assay in each tube type, 2000 ng/L BNP was spiked into each tube type evaluated that contained plasma collected from 4 healthy volunteers (<10 ng/L BNP), and BNP levels were quantified.

    Summary of Findings:

    Among the 36 patients evaluated, eight patients had <100 ng/L BNP in the EDTA/PET tube that was analyzed immediately, sixteen had a level between 100 and 500 ng/L BNP, and twelve had a level >500 ng/L BNP. BNP levels were significantly higher in plasma from EDTA PET tubes compared to EDTA glass tubes (mean 3.04%, P=0.015), and from EDTA glass tubes compared to lithium heparin glass tubes (mean 31. 4% higher, P < 0.001). BNP levels were unaffected by room temperature, post-centrifugation storage of plasma in EDTA/PET tubes but decreased slightly in EDTA/glass tubes (-0.73%, -0.66%, and -1.84% after 1, 2, and 4 h, respectively, relative to 30 min of storage). BNP levels decreased progressively when plasma was stored in lithium heparin glass tubes (-4.33%, -13.54%, and -26.62% after 1, 2, and 4 h, respectively, P < 0.001). When 2000 ng/L BNP was added to the plasma of healthy volunteers, 86.04% was recovered from EDTA glass tubes, 88.23% for EDTA PET tubes, but only 77.46% from lithium heparin glass tubes.
