Purpose of Paper
This paper reviewed the literature on the effects of room temperature storage of specimens on RNA integrity in eight tissue types.
Conclusion of Paper
Storage of lung, breast, ovary, prostate, pancreas, liver, and tonsil specimens at room temperature was found to have little to no effect on the integrity of RNA but was found to decline in 2 of 4 studies when colorectal tissue was stored at room temperature for 90-120 min.

Study Purpose
This study reviewed the literature on the effects of room temperature storage of specimens on RNA integrity in eight tissue types. Data from 14 papers investigating specimens from colorectal tumor and normal tissues, lung tumors, normal liver tissue, ovarian tumors, prostate tumor and normal tissue, breast tumor and normal tissues, normal tonsil, and pancreatic tumors were included. The number of patients studied ranged from 1-82. Details of specimen handling and analysis were not specified but it is assumed they were fresh and analyzed by bioanalyzer.
Summary of Findings:
With the exception of colorectal tissue, RNA integrity numbers (RIN) remained above 6 when specimens were stored at room temperature for up to 5 h. While RNA integrity in colorectal tissue as determined by RIN or the ratio of 28S to 18S rRNA was affected by 90-120 min at room temperature in two studies, the other two studies found no effect of 30 min and up to 16 h storage at room temperature on the ratio of 28S to 18S rRNA. Two studies demonstrated that storage at room temperature did not affect RNA integrity in lung (up to 2 h and 5 h, respectively), breast (3 h and 24 h, respectively) and ovary (2 h and 18 h, respectively) specimens. Further, the single studies investigating the effects of room temperature storage on pancreas and prostate found no effect on RIN after 1 h and a less than 15% decline in RIN after 8 h and a decrease in the 28S/18S rRNA ratio from 1.5 to 1.4 after 16 h on liver and tonsil specimens, respectively.
