Study Purpose
This study compared cell count, viability, and proliferative responsiveness of immune cells and subsets in freshly collected whole blood specimens, fresh Ficoll-isolated peripheral blood mononuclear cells (PBMC), and cryopreserved (frozen and thawed) PBMC. Biospecimens were collected from HIV-1 infected patients at nine participating institutions.
Summary of Findings:
Percentages of major lymphoctye subsets (T-cell, B-cell, CD19+) were significantly impacted by both fresh Ficoll-isolation of PBMC and cryopreservation of PBMC, while the percentage of CD4+ was elevated in freshly Ficoll-isolated PBMC compared to fresh whole blood and cryopreserved PBMC. Percentages of CD3+ and CD8+ did not differ among differentially processed specimens. A reduction in CD62L staining intensity, and a lower percentage of CD38+ and HLA-DR+ cells were observed in freshly Ficoll-isolated PBMC and those that were cryopreserved compared to fresh whole blood specimens. However, proliferative response was retained in differentially processed specimens, with a <10% probability of loss due to cryopreservation.
